Hexagon Bio Raises $62 Million and Advances HEX-360 to Clinical Trials
Hexagon Bio closed a $62 million Series C round led by The Column Group to initiate a Phase 1 trial for HEX-360 in the first half of 2027.

US-based Hexagon Bio announced the closing of a $62 million Series C funding round led by The Column Group, aiming to advance its drug HEX-360 into Phase 1 clinical trials.
The Menlo Park-headquartered company stated that the round saw participation from existing investors including Nextech Invest, Deviation Capital, 8VC, and Canada Pension Plan Investment Board (CPP Investments). It expects to initiate the Phase 1 trial for HEX-360 in the first half of 2027.
HEX-360 is an antibody-drug conjugate targeting the HER2 protein, carrying a novel translation-inhibitor payload—a mechanism distinct from approved payloads such as topoisomerase inhibitors. The company notes that cancer cells frequently develop resistance to circulating payloads, creating a need for novel mechanisms of action.
The funding will also be used to advance the company's platform to generate dual payloads and additional mechanisms of action. The company stated that this move reflects its conviction that the next generation of these therapeutics will need to overcome existing resistance mechanisms.
What do these terms mean?
Antibody-Drug Conjugate (ADC): A therapy that links a cancer-seeking antibody to a toxic payload, delivering the toxin to the target with higher precision and reducing the impact on healthy cells.
HER2: A protein present at high levels on the surface of certain cancer cells, used as a targeted marker for treatment in specific types of breast and gastric cancers.
Phase 1 Clinical Trial: The first test of a drug in humans, primarily aimed at assessing safety and appropriate dosage before evaluating efficacy on a broader scale.
